How is PDM 2009 H1N1 Different than the Original 2009 H1N1 ValuPanel Reagents?
Improved Specificity – the new probes and primers target more conserved regions of the same genes that were resolved in the original panel. They are therefore appropriate for detection of diverse isolates of Novel Influenza A H1N1 that emerged internationally, while discriminating against other flu types and strains.

In response to the outbreak of novel 2009 H1N1, the US Food and Drug Administration (FDA) issued an Emergency Use Authorization (EUA) to the CDC for the immediate use of the Real-time RT-PCR Detection Assays for 2009 H1N1 designed by the US Centers for Disease Control and Prevention (CDC). The CDC readily developed the 2009 H1N1 Panel to detect and identify cases of the novel influenza A (H1N1) virus, and is the first FDA-authorized distributor of this molecular diagnostic test to public health and other qualified laboratories. Biosearch Technologies participated in the rapid response to the Flu pandemic by supplying the probes and primers necessary for the H1N1 detection panel.

The master mix features high sensitivity and signal intensity as well as low background and better compatibility with cDNA templates derived directly from reverse transcription reaction mixture. The ExcelTaq™ 2X Q-PCR Master Mix (SYBR, no ROX) contains hot-start Taq polymerase in an optimized buffer with dsDNA specific SYBR green fluorescent dye. This master mix allows for sensitive, precise amplification, real-time tracking of the amplification process, and simultaneous quantification for targeted DNA molecules. With inert smart blue contrast dye, the ExcelTaq™ 2X Q-PCR Master Mix (SYBR, no ROX) is ready-to-use and greatly reduces pipetting errors, while largely improving the reproducibility of the process. Scorpions primers are probe/primer hybrids unlike other FRET probes, whose design is such that they emit light only when incorporated into the PCR product during amplification. These primers incorporate two distinct structures: 1) a target-specific DNA probing sequence and 2) a target-specific PCR primer.

The DNA probing element incorporates a self-complementary stem sequence at the 3' and 5' ends that holds the target specific region in a hairpin loop. A fluorophore molecule is covalently linked to the 5' end and, the dark quencher, a Black Hole Quencher dye, is linked to the 3' end of the loop structure. The fluorophore and BHQ dye are held in close physical proximity by the stem structure to ensure efficient quenching prior to thermal cycling.
